Global Analysis of Transcriptome Responses and Gene Expression Profiles to Cold Stress on Jatropha curca L.

Abstract:

Jatropha curcas L. (J. curcas), is an oil-rich shrub species with multiple uses especially for biodiesel production. Low temperature mainly affects their distribution and production. The newly developed Illumina Hiseq™ 2000 RNA-seq and Digital Gene Expression (DGE) are deep high-throughput approaches for gene expression analysis at the transcriptome level, by which we delicately investigated the gene expression profiles under cold stress in order to gain more insights into the molecular mechanism of cold response in J. curcas. 45,251 Unigenes were obtained by assembly of the clean data generated by RNA-seq analysis to the transcriptome of J. curcas. Among these Unigenes, 4185 members are possibly associated with cold-resistance. Changes in gene expression were evaluated by DGE analysis for 12°C cold exposure of 12h, 24h, 48h time periods. The results revealed a total of 3178 Unigenes significantly up-regulated for expression and 1244 down-regulated under such cold stress. In addition, 239, 160, 333 novel genes of J. curcas were induced after 12h, 24h, 48h of 12°C treatment, respectively. These results not only help improve the current understanding of plant cold-resistance mechanism, but also can favor the screening of crucial genes for genetic improvement on the cold resistance in J. curcas.
