Abstract:

Glycerol, a substantial byproduct of conventional biodiesel synthesis, is a promising carbon source for microbial transformation into more valuable biochemicals. Butanol, an important intermediate and holds potential as a renewable energy source with fuel properties similar to gasoline, could be synthesized from acetone-butanol-ethanol (ABE) fermentation using Clostridium sp. This study investigated the utilization of glycerol, the biodiesel byproduct, as the sole carbon source. Initial screening confirmed three Clostridium sp. isolates could produce ABE from pure glycerol. The C13 isolate was selected for optimization after achieving the highest butanol concentration (2.66 ±0.17 g/L) in T6 medium after 72 hours. Subsequently, the C13 isolate was evaluated across five media formulations (MP2, Lin, Semi-defined, Anaerobic Sugar (AnS), and T6) to maximize butanol yield. The results indicated that the T6 medium yielded the maximum acetone (0.56 ±0.09 g/L), butanol (3.26 ±0.30 g/L) and ethanol (2.96 ±0.54 g/L) concentrations at 96 h. The Anaerobic Sugar (AnS) medium provided the highest ethanol production (3.77 ±0.49 g/L at 72 h), while no ABE production was observed in the MP2 medium.
