Cellulase Production by Fungi from Cassava Stems Applied to the Hydrolysis of Sugarcane Bagasse Treated with Ionic Liquid for Bioethanol Production

Abstract:

In this work, it was evaluated the cellulase production by 14 fungi isolated from cassava stems (Microdochium lycopodinum, Colletotrichum sp, Cladosporium sp., Alternaria sp, Phomopsis sp, Diaporthe phaseolorum, Diaporthe endophytica, Vouchered mycorrhizae, Phanerochaete australis, Diaporthe caatingaensis, Stenocarpella maydis, Annulohypoxylon stygium, Sordariomycetes sp, Colletotrichum gloeosporioides, and Phanerochaetaceae sp.). It was possible to verify a higher production of cellulases by the Annulohypoxylon stygium (0.83 ± 0.01 mg.mL-1 of fermentable sugars). The influence of time in the growth of this fungus was analyzed and it was possible to determine an ideal growth period of 13 days (enzymatic activity of 0.212 FPU.L-1.min-1). The ideal growth pH was also evaluated for this fungus, being verified in pH 4 the activity of 0.427 ± 0.003 FPU.mL-1.min-1. The enzymatic extract was pre-purified using an ammonium sulphate precipitation and presented a higher cellulase activity in the fraction obtained after 50 % of saturation (0.430 ± 0.002 FPU.mL-1). In addition, the efficiency of enzymatic hydrolysis of sugarcane bagasse treated with sec-butylammonium acetate IL was evaluated with the enzymatic extract obtained from the Annulohypoxylon stygium and the ideal hydrolysis time was evaluated in 32 h (FPU=1.345 ± 0.039 mg.mL-1.min-1).
