Lignin and Copper Increase Rice Husks Delignification in the Enzymatic Pretreatment with Culture Extracts from Pleurotus Ostretus

Abstract:

In this study, the pretreatment of rice husks with enzymatic extracts produced by Pleurotus ostreatus was evaluated. Two culture media compositions to produce enzymatic extracts were chosen from sixteen culture media with glucose, and yeast-extract supplemented with carboxymethylcellulose (CMC), xylose, and lignin-alkali, with or without the laccase inducer, copper sulfate, which were initially for laccase activity and laccase isoenzymes. The extracts produced with lignin had laccase activity, total protein concentration, and laccase isoenzymes different than the other extracts produced with CMC or xylose or both. Therefore, the compositional, chemical, and structural characterization of the rice husks treated with these extracts produced with lignin and with or without copper was compared. The results showed a lignin loss between 24 and 37%, changes structural on spectra FTIR (Fourier Transform Infrared Spectrometry) on the region between wavenumbers 1740 and 1512 cm-1 which are mainly assigned to C=O stretching, and C=C stretching by aromatic skeletal. The morphological modification as seen SEM (Scanning Electronic Microscopic) showed changes in the rice husk internal layer. These results suggest that the lignin modification by the enzymatic P. ostreatus metabolism natural produce lignin derived that could act as laccase-mediator increase their delignification power.
