Goat and Sheep Cheese Whey by Products Can Be Used for the Production of Recombinant Proteins from Escherichia Coli

Abstract:

Cheese whey (CW) is one of the major dairy by products. Although it contains soluble proteins, lactose, lipids, mineral salts, and vitamins, it is usually disposed of becoming thus a pollutant. We examined whether mixed CW from goat and sheep could be used as an alternative growth medium to produce recombinant proteins from Escherichia coli (E. coli). In a first step, the mixed goat and sheep CW was analyzed to determine possible nutritional sources for bacterial growth. Various sugars (glucose, lactose, galactose), organic acids (lactic acid, succinic acid), and ethanol were detected by HPLC, while total nitrogen content was estimated by the Kjeldahl method, and total proteins by the method of Bradford. Although nutrients were plentiful for the growth of bacteria, the very high turbidity of CW did not allow for easy measurements of bacterial growth. By pretreating with ZnSO4 and continuing with the EP0064509B1 patent protocol, significant clearing of CW was achieved. The resulting medium allowed for the growth of E. coli and the expression of E. coli thioredoxin 1 and thioredoxin reductase from Plasmodium falciparum. Expression yields (SDS-PAGE) were comparable to those from the LB medium.
