Optimization of Extraction of Bioactives from Different Wastes and By-Products of Agro- and Food Industry

Abstract:

We aimed to quantify the bioactives (natural preservatives and functional substances) from agro- and food industrial waste streams and offer technological solutions to produce value-added products from agro-food wastes. Extracts with potent antioxidant properties were obtained from lavender steam- and hydro-distilled residues, sea buckthorn stalks and from swept walnut tree- and freshly collected black mulberry leaves. The applied methods (Soxhlet-, stirred-, and ultrasound assisted extraction), solvents, and process parameters of extraction (extraction time, temperature and feed-to-solvent ratio) were optimized for all waste sources aiming for the highest extraction yield with the highest concentration of the targeted bioactives in the extracts. The antioxidant activity of extracts was analyzed by the DPPH method in the presence of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical. The content of total phenolics was quantified using Folin-Ciocalteu’s phenol reagent. Individual phenolic compounds were quantified by HPLC and LC-MS/MS. The antioxidant activities correlate well with the total polyphenol contents. The extracts of sea buckthorn stalk, lavender steam distilled residue and walnut leaves showed as strong antioxidant activities as the synthetic alternatives (BHT, BHA). Extracts from sea buckthorn stalk exhibited strong antibacterial activities in standard in vitro antimicrobial tests in the presences of different strains of Gram-negative and Gram-positive bacteria.
